Model-guided design of ligand-regulated RNAi for programmable control of gene expression
A general approach for assembling shRNA switches from modular functional components is described, providing programmable ligand-controlled gene regulatory elements in mammalian cells. A first-generation modeling framework is demonstrated that predicts sequence-to-function relationships and guides the forward engineering of shRNA switch properties.
